Q1: What are the main stages of the zebrafish life cycle?
The zebrafish lifecycle consists of four major periods: embryo, larva, juvenile, and adult. Development from fertilized egg to adult takes approximately 90 days. Early development occurs at a rapid, predictable rate when embryos are raised at 28°C, allowing researchers to define stages by hours or days post fertilization (hpf or dpf).
Q2: What happens during the first 24 hours of zebrafish development?
The first 24 hours are divided into five phases: zygote, cleavage, blastula, gastrula, and segmentation. During cleavage, the blastodisc divides into blastomeres that undergo rapid cell divisions. The blastula period begins when the embryo makes its own RNA. Gastrulation produces three germ layers—endoderm, mesoderm, and ectoderm—that give rise to different tissues.
Q3: How do the three germ layers contribute to zebrafish organ formation?
The ectoderm gives rise to epidermis and the nervous system, the endoderm forms the gut, and the mesoderm generates muscle, bone, and vasculature. By 12 hours post fertilization, the mesoderm begins dividing into somites, which are tissue segments along the trunk that later become muscle. These distinct cell layers establish the basic body plan.
Q4: What structural features make zebrafish embryos ideal for developmental research?
Zebrafish embryos are transparent, allowing direct visualization of tissue morphogenesis at the cellular level. Their rapid, external development and amenability to both physical and genetic manipulations enable researchers to study developmental processes. Additionally, zebrafish have high genetic similarity to humans, making them valuable for understanding vertebrate development and disease.
Q5: How are zebrafish larvae raised from hatching to adulthood?
After hatching at approximately 3 days post fertilization, larvae are placed in tanks with minimal water exchange initially, with increasing water flow as fish improve their swimming and hunting abilities. Larvae are fed rehydrated dry food and microorganisms like paramecia to maximize growth. After 2-3 months in the nursery, young fish reach adulthood, completing the lifecycle.
Q6: What techniques are used to study gene expression and cell movement in zebrafish embryos?
RNA in situ hybridization uses labeled RNA molecules to visualize gene expression throughout the organism, revealing changes over time and in specific organs. Cell transplantation involves transferring fluorescently labeled cells into host embryos to track movements and examine cell interactions. Microinjection microinjection early zebrafish embryos allows researchers to examine specific gene roles by injecting morpholinos to block protein expression.
Q7: Why are morpholino injections useful for studying zebrafish development?
Morpholinos are antisense oligonucleotides designed to block protein expression, allowing researchers to study loss-of-function effects of specific genes starting from the one-cell stage. By screening injected fish for developmental phenotypes, such as changes in blood vessel assembly, scientists gain insight into the genetic control of complex developmental processes in a vertebrate model.
