B Cell Production

B cell production is the biological process through which hematopoietic stem cells generate, mature, and maintain B lymphocytes, the immune cells responsible for antibody-mediated defense. In mammals, precursor cells develop primarily in the bone marrow, where sequential differentiation and V(D)J gene rearrangement create unique B cell receptors; developing cells undergo selection to remove or revise strongly self-reactive receptors. Mature naïve B cells then migrate to peripheral lymphoid organs, where antigen recognition, with appropriate signaling, can drive their formation into antibody-secreting plasma cells or long-lived memory cells. Understanding this process supports research into immune development, vaccination, immunodeficiency, autoimmunity, and B cell malignancies.

In this protocol we describe production, purification and titration of lentiviral vectors. We provide an example of lentiviral vector-mediated gene delivery in primary cultured neurons and astrocytes. Our methods may also apply to other cell types in vitro and in vivo.

An efficient system of structure and function analysis of a gene in an ex vivo culture of splenic B-lymphocytes is described. This method takes advantage of recombinant retroviral production in a helper free, ecotrophic packaging cell line. Stable, heritable expression of a gene of interest within primary lymphocytes is achieved leading to generation of surface antibodies on B cells undergoing class switch recombination.
