Beta-catenin Gain-of-function

Beta-catenin gain-of-function refers to increased or constitutive activity of beta-catenin, a protein that regulates cell adhesion and Wnt-dependent gene expression. Normally, the destruction complex phosphorylates cytoplasmic beta-catenin and targets it for degradation; activating mutations or altered signaling can prevent this process, allowing beta-catenin to accumulate, enter the nucleus, and activate TCF/LEF target genes. This mechanism influences cell fate, proliferation, and tissue patterning during development. Studying beta-catenin gain-of-function helps researchers model developmental abnormalities, investigate tumor formation, and evaluate how dysregulated Wnt signaling contributes to disease.

Catenins are characterized by multiple binding domains and dynamic structures that allow them to function as linker proteins in cell junction complexes. All catenins, except α-catenin, contain a characteristic protein sequence called the armadillo repeat and are therefore also called armadillo proteins. Catenins in Cell Junctions Catenins bind to cell adhesion molecules such as cadherins and link them to different cytoskeletal proteins depending on the type of cell junction. At the adherens...

This protocol allows one to identify factors that modulate functional beta cell mass to find potential therapeutic targets for the treatment of diabetes. The protocol consists of a streamlined method to assess islet replication and beta cell function in isolated rat islets following manipulation of gene expression with adenoviruses.

Gain and phase shift are properties of linear circuits that describe the effect a circuit has on a sinusoidal input voltage or current. The circuit's behavior that contains reactive elements will depend on the frequency of the input sinusoid. As a result, it is observed that the gain and phase shift will all be frequency functions. Gain: Suppose Vin is the input and Vout is the output signal to a circuit. The input and output signals have the amplitude A and B and a phase angle of 0° and θ°,...

In this article, we present two complementary methods to measure specific rates of translational error and mistranslation in the model mycobacterium, Mycobacterium smegmatis, using gain-of-function reporter systems. The methods can be employed to measure accurate error rates in low throughput or relative error rates in a more high-throughput setting.
