Lung Fixation

Lung fixation is the preservation of lung tissue in a stable state so its architecture, cellular features, and disease-related changes can be examined. In cancer research, fixatives such as formalin chemically cross-link proteins and other cellular components, while controlled inflation or intratracheal instillation helps maintain the lung’s air-space structure during processing. Fixed tissue can then be sectioned, stained, and analyzed by histology, immunohistochemistry, or molecular assays to assess tumor development, invasion, and responses to treatment. Consistent fixation improves tissue comparability across experiments and supports accurate evaluation of preclinical lung cancer models.

Presented is a method for air-inflation with vascular perfusion-fixation of the lungs that preserves the location of cells within airways, alveoli and interstitium for structure-function analyses. Constant airway pressure is maintained with an air-inflation chamber while fixative is perfused via the right ventricle. Lungs are processed for histologic studies.

Presented here is a useful protocol for lung fixation that creates a stable condition for histological evaluate of lung specimens from a mouse model of emphysema. The main advantage of this model is that it can fix many lungs with the same constant pressure without lung collapse or deflation.
