Tumor Establishment

Tumor establishment is the process by which transformed cells initiate, survive, and form a persistent mass within a tissue, making it a central concept in cancer research. It begins when genetic or epigenetic alterations disrupt growth control, while interactions with the extracellular matrix, blood vessels, and immune cells help tumor cells evade elimination and secure nutrients. Studying these early stages clarifies how cancers adapt to their microenvironment and progress from isolated abnormal cells to organized tumors. Experimental models of tumor establishment support research on cancer initiation, metastasis, biomarker development, and therapies designed to prevent tumor growth or disrupt its supporting niche.

Luciferase-modified human brain tumor xenografts can be established intracranially in athymic mice, with subsequent monitoring of tumor growth and response to therapy using bioluminescence imaging. In combination with survival analysis, bioluminescence monitoring is an essential research tool for pre-clinical testing of therapies being considered for treating brain tumors.

Establishment and Propagation of Human Retinoblastoma Tumors in Immune Deficient Mice

A method is described to propagate human retinoblastoma tumors in mice. Tumor cells are directly injected into the eyes of immune deficient mice. Secondary tumors have been successfully established using both cells directly harvested from human tumors and cultured tumorspheres.
