Tissue Differentiation

Tissue differentiation is the developmental process by which unspecialized cells acquire distinct identities, functions, and structures, enabling the formation of organized tissues and organs. It occurs when extracellular signals activate specific transcription factors and patterns of gene expression, while cell-cell interactions and the extracellular matrix reinforce lineage decisions and spatial organization. In developmental biology, studying tissue differentiation clarifies how embryos establish functional systems and how disruptions can lead to congenital disorders or disease. These principles also support research in stem cell differentiation, regenerative medicine, organoid development, and tissue engineering, where scientists aim to generate specialized cells and construct functional replacement tissues.

Isolation and Differentiation of Adipose-Derived Stem Cells from Porcine Subcutaneous Adipose Tissues

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.

In this video, we describe a technique of hematoxylin and eosin staining of paraffin-embedded tissue sections to study their histology. The procedure includes deparaffinization of the tissue followed by staining the cells in the tissue. Upon staining, the nucleus appears distinctly blue, and the proteins in the cytoplasm display pink coloration.

Rapid Isolation And Purification Of Mitochondria For Transplantation By Tissue Dissociation And Differential Filtration

A method for rapid isolation of mitochondria from mammalian tissue biopsies is described. Rat liver or skeletal muscle preparations were homogenized with a commercial tissue dissociator and mitochondria were isolated by differential filtration through nylon mesh filters. Mitochondrial isolation time is <30 min compared to 60 - 100 min using alternative methods.
