Allogeneic Hsct

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) replaces a patient’s blood-forming stem cells with cells from a genetically compatible donor, offering potentially curative treatment for hematologic cancers, marrow failure, and selected immune disorders. Before infusion, conditioning therapy suppresses the recipient’s bone marrow and immune system; donor stem cells then engraft and reconstitute blood and immune cell production, while donor T cells may attack residual malignant cells through the graft-versus-leukemia effect. In immunology and infection research, allo-HSCT illustrates immune reconstitution, histocompatibility, graft-versus-host disease, and susceptibility to opportunistic infections, guiding donor selection, prophylaxis, monitoring, and post-transplant care.

Tail-skin transplantation is a powerful model for studying T cell-dependent rejection and tolerance induction during allogeneic immune responses in mice. The advantages of this protocol are minor invasive surgery, and ease of monitoring with no need to sacrifice the recipient mouse.

This manufacturing process is GMP-compliant and should ensure the broader implementation of T-cell immunotherapy beyond specialized centers.
