Cerebral Oxygenation

Cerebral oxygenation is the delivery and utilization of oxygen within brain tissue, a process essential for neuronal metabolism and normal neurological function. It depends on cerebral blood flow, arterial oxygen content, and diffusion into tissue; because oxygenated and deoxygenated hemoglobin absorb near-infrared light differently, near-infrared spectroscopy can estimate regional tissue oxygenation noninvasively. In medicine, cerebral oxygenation assessment supports monitoring during anesthesia and surgery and helps evaluate patients with stroke, traumatic brain injury, neonatal complications, or impaired cerebral perfusion. Tracking changes can reveal inadequate oxygen supply early, guide clinical management, and improve understanding of brain physiology and injury.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

We combined frequency-domain near-infrared spectroscopy measures of cerebral hemoglobin oxygenation with diffuse correlation spectroscopy measures of cerebral blood flow index to estimate an index of oxygen metabolism. We tested the utility of this measure as a bedside screening tool to evaluate the health and development of the newborn brain.

2011

Surgical induction of ischemic brain damage in the rat is a widely used model for stroke research. Here we demonstrate the induction of focal cerebral ischemia by occlusion of the middle cerebral artery. Visualization of the resulting infarct by histological staining and magnetic resonance imaging is also shown.

Detecting Reactive Oxygen Species

2023

Reactive oxygen species are chemically active, oxygen-derived molecules capable of oxidizing other molecules. Because of their reactive nature, there are many deleterious effects associated with unchecked ROS production, including structural damage to DNA and other biological molecules. However, ROS can also be mediators of physiological signaling. There is accumulating evidence that ROS play significant roles in everything from activation of transcription factors to the mediation of...

Induction of Cerebral Microhemorrhages in a Rat Model Using Lipopolysaccharide

2025

This video demonstrates the process of inducing blood-brain barrier (BBB) disruption and cerebral microhemorrhages in a rat model using lipopolysaccharide (LPS). It highlights the sequence of immune responses that lead to increased BBB permeability and subsequent brain tissue damage.
