Focal Brain Ischemia

Focal brain ischemia is a localized reduction or interruption of blood flow to part of the brain, depriving affected tissue of oxygen and glucose and potentially causing neurological injury. It commonly results from an arterial obstruction, which disrupts energy production, impairs ion balance, promotes excitotoxicity, and can lead to irreversible infarction if perfusion is not restored. In medicine, focal brain ischemia is central to understanding ischemic stroke, its clinical manifestations, and the distinction between salvageable and permanently damaged tissue. Experimental models and imaging methods help researchers evaluate injury mechanisms, treatment timing, neuroprotection, and recovery after cerebrovascular events.

There is accumulating evidence, that ischemic preconditioning (PC) – a non-damaging ischemic challenge to the brain - confers a transient protection to a subsequent damaging ischemic insult. We established bilateral common carotid artery occlusion (BCCAO) as a preconditioning stimulus to induce early ischemic tolerance (IT) to transient focal cerebral ischemia (induced by middle cerebral artery occlusion, MCAO) in C57Bl6/J mice.

Focal Cerebral Ischemia Model by Endovascular Suture Occlusion of the Middle Cerebral Artery in the Rat

Surgical induction of ischemic brain damage in the rat is a widely used model for stroke research. Here we demonstrate the induction of focal cerebral ischemia by occlusion of the middle cerebral artery. Visualization of the resulting infarct by histological staining and magnetic resonance imaging is also shown.

Bilateral carotid occlusion coupled with systemic hypotension produces global brain ischemia in the rat, resulting in damage to the hippocampus with reproducible severity. Animal subjects are impaired with predictable patterns of brain damage, they recover expediently, and mortality rates are comparatively low.
