Cholesterol Rich Liposomes

Cholesterol-rich liposomes are lipid vesicles containing a high proportion of cholesterol, a membrane component that helps tune bilayer structure and performance for bioengineering applications. Cholesterol inserts between phospholipids, influencing molecular packing, membrane fluidity, permeability, and resistance to disruption, while the aqueous core and lipid bilayer provide compartments for carrying soluble or membrane-associated compounds. These properties support the design of drug-delivery systems, biomimetic membranes, and platforms for studying membrane interactions. By adjusting cholesterol content alongside lipid composition and preparation conditions, researchers can control vesicle stability, cargo retention, and interactions with biological environments, improving the functional design of engineered delivery systems.

The goal of this protocol is to use cationic/anionic liposomes with a neuro-targeting peptide as a CNS delivery system to enable siRNA to cross the BBB. The optimization of a delivery system for treatments, like siRNA, would allow for more treatment options for prion and other neurodegenerative diseases.

In this article, a protocol for infection of macrophages with Cryptococcus neoformans is described. Also, a method for sterol depletion from the macrophages is explained. These protocols provide a guide to study fungal infections in vitro and examine the role of sterols in such infections.

Reconstituting functional membrane proteins into giant liposomes of defined composition is a powerful approach when combined with patch-clamp electrophysiology. However, conventional giant liposome production may be incompatible with protein stability. We describe protocols for producing giant liposomes from pure lipids or small liposomes containing ion channels.

The cholesterol assay is designed to quantitate the rate of cholesterol efflux from cultured cells and the capacity of plasma acceptors to accept cholesterol released from cells.
