Apoptotic Foci

Apoptotic foci are discrete cellular or tissue regions in which programmed cell death is concentrated or detected, providing spatial evidence of apoptosis in biological samples. They arise as apoptotic signaling activates initiator and executioner caspases, leading to cellular shrinkage, chromatin condensation, DNA fragmentation, and formation of membrane-bound apoptotic bodies that can appear as localized microscopic signals. Identifying these foci through fluorescence microscopy, tissue staining, or molecular markers helps researchers map cell loss, quantify treatment responses, and study development, disease progression, and tissue remodeling. Their distribution and abundance can reveal where and when apoptosis occurs within complex biological systems.

Quantitation of DNA double-strand breaks on the basis of γH2AX foci has become an invaluable tool, particularly in radiation biology, for the evaluation of tissue radiosensitivity and effects of radiation modifying compounds. Here we demonstrate the use of an immunofluorescence assay for quantitation of γH2AX foci in tissue samples.
