Immunohistochemistry

Immunohistochemistry (IHC) is a laboratory method that uses antibodies to detect and locate specific proteins, or other antigens, within preserved tissue sections, linking molecular identity to tissue structure. In the procedure, a primary antibody binds its target, while an enzyme-linked or fluorescent detection system produces a visible signal that can be examined by light or fluorescence microscopy. IHC helps researchers map protein distribution, characterize cell types, and investigate tissue organization, disease-associated changes, and biological pathways; in clinical settings, standardized staining patterns can also support tumor classification and diagnosis. Careful controls and optimized fixation are essential for reliable interpretation.

Immunocytochemistry (ICC) and immunohistochemistry (IHC) are techniques that use antibodies to check for specific proteins or antigens in a sample. The technique was first published by Albert Coons in 1941 to detect the presence of pneumococcal antigen in tissue sections from mice infected with Pneumococcus. Immunocytochemistry helps localization of proteins or antigens in individual cells like blood cells, stem cells, etc., while immunohistochemistry does the same for tissue samples. These...

Neural circuits are topographically organized into functional compartments with specific molecular profiles. Here, we provide the practical and technical steps for revealing global brain topography using a versatile wholemount immunohistochemical staining approach. We demonstrate the utility of the method using the well-understood cytoarchitecture and circuitry of cerebellum.
