Murine Liver

Murine liver refers to the liver of mice, a well-characterized organ used to study metabolism, regeneration, disease, and cancer biology. Its hepatocytes are organized into lobules supplied by sinusoids, where blood-borne nutrients, signaling molecules, and compounds undergo processing that can influence tumor initiation and progression. In cancer research, researchers use genetically engineered mice, chemically induced tumors, and transplantation models to examine hepatocarcinogenesis, tumor–microenvironment interactions, therapeutic response, and metastasis. These models connect cellular and molecular findings with whole-organ physiology, although differences between mouse and human liver biology must be considered when interpreting translational relevance.

Here we describe a common method to induce chronic liver injury in mice by feeding of a choline-deficient and ethionine-supplemented (CDE) diet. We demonstrate health monitoring, liver perfusion, isolation, and preservation. A time course of six weeks can inform about liver injury, pathohistology, fibrosis, inflammatory, and liver progenitor cell responses.

Murine Precision-Cut Liver Slices as an Ex Vivo Model of Liver Biology

This protocol provides a simple and reliable method for the production of viable precision-cut liver slices from mice. The ex vivo tissue samples can be maintained under laboratory tissue culture conditions for multiple days, providing a flexible model to examine liver pathobiology.

A high incidence of tumor recurrence after resection of liver metastases remains an unsolved problem. The illustrated mouse model may be useful to investigate the reasons for such recurrences. It combines a liver resection model with intrahepatic tumor cell injection for the first time.
