Bfu-e Progenitors

BFU-E progenitors, or burst-forming unit-erythroid progenitors, are early hematopoietic cells that generate erythroid lineages and help reveal how red blood cells develop. In semisolid culture, individual progenitors respond to erythropoietin and cooperating growth signals, undergo several rounds of proliferation, and form large, multicellular colonies that mature into erythroblasts. Their abundance, colony-forming capacity, and progression toward red blood cell production provide measurable indicators of erythropoiesis. In developmental biology, BFU-E assays help characterize blood formation, compare progenitor potential across tissues or developmental stages, and assess how genetic changes, signaling pathways, or external factors influence erythroid differentiation.

Differentiation of Neural Progenitor Cells into Neurons

The video showcased a cell culture method for differentiating neural progenitor cells into neurons. By incubating cells in a nutrient-rich medium supplemented with growth factors, the progenitor cells matured into neurons with developed axons and dendrites. A stabilizing agent ensured cell viability and protected against oxidative damage throughout the differentiation process.

Primary, human fetal brain-derived, multipotential progenitor cells proliferate in vitro while maintaining the capacity to differentiate into neurons and astrocytes. This work shows that neural progenitors can be induced to differentiate through stages of the oligodendrocytic lineage by conditioning with select growth factors.

A method to analyse the distribution of bone marrow hematopoietic progenitors in flow cytometry as well as to efficiently isolate highly purified hematopoietic stem cells (HSCs) is described. The isolation procedure is essentially based on magnetic enrichment of c-Kit+ cells and cell sorting to purify HSCs for cellular and molecular studies.

While infiltrating macrophages are continuously recruited to adult tissues from circulating precursors, resident macrophages seed their tissue during development, where they are maintained without further input from progenitors. The progenitors for resident macrophages were recently identified. Here, we present methods for the genetic fate mapping of the resident macrophage progenitors.
