Endometrial Epithelial Cells

Endometrial epithelial cells are the specialized cells lining the uterus, where they form a dynamic barrier and contribute to the tissue’s preparation for pregnancy. Their behavior changes in response to ovarian hormones: estrogen promotes proliferation, while progesterone supports differentiation and the secretory functions required for embryo implantation. In developmental biology, these cells provide a model for studying epithelial renewal, cell signaling, tissue remodeling, and interactions between the maternal lining and the developing embryo. Primary cultures, organoids, and engineered endometrial tissues help investigate implantation, infertility, menstrual disorders, and mechanisms of reproductive tract development.

We present a protocol that combines cell isolation and whole-cell patch-clamp recording to measure the electrical properties of the primary dissociated epithelial cells from the rat cauda epididymides. This protocol allows for investigation of the functional properties of primary epididymal epithelial cells to further elucidate the physiological role of the epididymis.
