Infection Protocol

An infection protocol is a standardized, controlled plan for introducing an infectious agent into a biological system and measuring the resulting infection and host response. It specifies critical variables such as the agent or strain, inoculum, exposure route, timing, containment conditions, sampling schedule, and analytical methods, enabling consistent and interpretable experiments. In immunology and infection research, these protocols support investigation of pathogen replication, innate and adaptive immune responses, inflammation, and disease progression. Carefully designed procedures improve reproducibility, protect researchers and study subjects, and provide a foundation for evaluating vaccines, therapeutics, diagnostic methods, and host-directed interventions.

Once a gene is identified as potentially refractory for the dengue virus, it must be evaluated for it's role in preventing viral infections within the mosquito. This protocol illustrates how the extent of dengue infections of mosquitoes can be assayed. The techniques for growing up the virus in culture, membrane feeding mosquitoes human blood, and assaying viral titers in the mosquito midgut are demonstrated.

C. elegans has emerged as a new genetic model to study host-pathogen interactions. Here we describe a protocol to infect C. elegans with Salmonella typhimurium coupled with the double-strand RNAi interference technique to examine the role of host genes in defense against Salmonella infection.
