Isothermal Amplification

Isothermal amplification is a nucleic acid amplification technique that generates many copies of DNA or RNA-derived targets at a constant temperature, offering a simpler alternative to temperature-cycling methods such as PCR. It uses specialized primers and strand-displacing polymerases, or related enzymes, to repeatedly separate and copy target sequences without thermal cycling. In immunology and infection research, methods such as loop-mediated isothermal amplification support rapid detection of bacteria, viruses, and other pathogens from clinical or environmental samples. Their speed, sensitivity, and minimal equipment requirements can enable point-of-care testing, outbreak surveillance, and diagnostic workflows in settings with limited laboratory infrastructure.

Diagnosis of Eimeria infection in chickens remains demanding. Parasite morphology- and host pathology-led approaches are commonly inconclusive, while molecular approaches based on PCR have proven demanding in cost and expertise. The aim of this protocol is to establish loop-mediated isothermal amplification (LAMP) as a straightforward molecular diagnostic for eimerian infection.

This paper reports the protocol for a rapid identification assay for Bemisia tabaci based on loop-mediated isothermal amplification (LAMP) technology. The protocol requires minimal laboratory training and can, therefore, be implemented on-site at points of entry for plant imports such as seaports and airports.
