Chronic Implantation Rats

Chronic implantation rats are laboratory rats fitted with devices that remain in place after recovery, allowing repeated access to neural tissue for neuroscience experiments without performing a new procedure each time. During implantation, electrodes, cannulas, or other probes are secured to the skull and connected to recording or stimulation systems; subsequent measurements can be collected in awake, behaving animals over extended periods. This preparation supports longitudinal studies of neural activity, circuit function, behavior, and responses to experimental stimulation or pharmacological manipulation, while reducing variability associated with repeated surgeries. Careful surgical technique, postoperative monitoring, and stable device placement are essential for reliable data and animal welfare.

Source: Sanchez, C. A., et al. Preparation of Peripheral Nerve Stimulation Electrodes for Chronic Implantation in Rats. J. Vis. Exp. (2020).This video demonstrates vagus nerve stimulation in an awake rat. A rat with an implanted chronic peripheral nerve cuff electrode and trained in a lever press task is connected to a stimulus generator via an implanted head cap. The vagus nerve is stimulated immediately after each successful lever press, resulting in the expansion of the motor map.

Existing approaches for constructing chronically implantable peripheral nerve cuff electrodes for use in small rodents often require specialized equipment and/or highly trained personnel. In this protocol we demonstrate a simple, low-cost approach for fabricating chronically implantable cuff electrodes, and demonstrate their effectiveness for vagus nerve stimulation (VNS) in rats.
