Postnatal Rat Brain

The postnatal rat brain is the brain tissue of rats during the period after birth, when neural circuits, cell populations, and supporting structures continue to mature. During this stage, neurogenesis in selected regions, synaptic formation and pruning, myelination, and changes in gene expression shape connectivity and function in response to developmental signals and experience. Neuroscience researchers study postnatal rat brains to examine brain development, plasticity, neurodegenerative processes, injury responses, and the effects of drugs or environmental conditions. Analyses of tissue structure, molecular markers, electrophysiology, and behavior can link cellular changes to emerging neural functions and inform broader mammalian research.

Neuroblast migration is a fundamental event in postnatal neurogenesis. We describe a protocol for efficient labeling of neuroblasts by in vivo postnatal electroporation and subsequent visualization of their migration using time-lapse imaging of acute brain slices. We include a description for the quantitative analysis of neuroblast dynamics by video tracking.

Mosaic Analysis of Gene Function in Postnatal Mouse Brain Development by Using Virus-based Cre Recombination

An in vivo method to test gene function in postnatal brain is described. Recombinant AAVs expressing Cre and/or a fluorescent protein are injected into neonatal mouse brain. Mosaic gene inactivation and sparse neuronal labeling are achieved, allowing rapid analysis of gene function in processes critical to neural circuit development.

Stereotaxic Surgery for Excitotoxic Lesion of Specific Brain Areas in the Adult Rat

Targeted ablation of specific brain region(s) by infusion of an excitotoxin using stereotaxic coordinates is described. This technique could also be adapted for infusion of other chemicals into the rat brain.
