3d Localization Microscopy

3D localization microscopy is a super-resolution imaging approach that determines the three-dimensional positions of individual fluorescent molecules, revealing molecular organization beyond the diffraction limit of conventional light microscopy. It works by detecting sparse, temporally separated fluorescence signals and estimating each molecule’s position from its point-spread function, with axial information obtained through methods such as astigmatism or engineered point-spread patterns. In biochemistry, this technique maps protein distributions, molecular interactions, and dynamic assemblies within cells and biomolecular systems. Its nanometer-scale measurements help connect biochemical function with spatial organization and support studies of complex cellular mechanisms.

Nano-fEM: Protein Localization Using Photo-activated Localization Microscopy and Electron Microscopy

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.

Correlative Microscopy for 3D Structural Analysis of Dynamic Interactions

We describe a correlative microscopy method that combines high-speed 3D live-cell fluorescent light microscopy and high-resolution cryo-electron tomography. We demonstrate the capability of the correlative method by imaging dynamic, small HIV-1 particles interacting with host HeLa cells.

Ultrasound Localization Microscopy for Super-Resolution Mapping of the Rodent Brain Microvasculature

Ultrasound Localization Microscopy (ULM) is a recently developed technique that allows for an unprecedented level of resolution in imaging the microvasculature in vivo. In this work, we describe in detail how to obtain super-resolved images of the brain microvasculature in rodents using a standardized functional ultrasound imaging platform designed for preclinical research.
