Immune Cell Enrichment

Immune cell enrichment is the selective isolation or concentration of specific immune cell populations from complex biological samples, enabling more precise analysis of immune function in cancer research. It typically relies on differences in cell-surface markers, physical properties, or density, using approaches such as magnetic-activated cell sorting, flow-based separation, or density-gradient centrifugation. Enriched populations may include T cells, B cells, natural killer cells, or tumor-infiltrating lymphocytes obtained from blood or tumor tissue. Researchers use these cells to characterize the tumor immune microenvironment, measure responses to treatment, and support studies of cancer immunotherapies and biomarker development.

Studying Organelle Dynamics in B Cells During Immune Synapse Formation

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.

This protocol describes the use of peptide:MHC tetramers and magnetic microbeads to isolate low frequency populations of epitope-specific T cells and analyze them by flow cytometry. This method enables the direct study of endogenous T cell populations of interest from in vivo experimental systems.

Isolation of Functional Cardiac Immune Cells

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

Here we describe a method to separate and enrich components of the tumor immune and non-immune microenvironment in established subcutaneous tumors. This technique allows for the separate analysis of tumor immune infiltrate and non-immune tumor fractions which can permit comprehensive characterization of the tumor immune microenvironment.
