Fungal Mutant Screening

Fungal mutant screening is a method for identifying fungal strains with genetic changes that alter observable traits, helping researchers link genes to cellular and developmental functions. Researchers generate or obtain mutant populations, grow them under controlled conditions, and compare phenotypes such as growth, morphology, sporulation, or formation of specialized structures with those of a reference strain. By associating a phenotype with a disrupted or modified gene, then confirming the relationship through genetic analysis or complementation, screening reveals mechanisms that regulate fungal development. In developmental biology, this approach supports studies of cell differentiation, multicellular patterning, signaling, and environmental responses, while also informing research on pathogenicity, metabolism, and biotechnology.

High Throughput Screening of Fungal Endoglucanase Activity in Escherichia coli

We describe a low cost, high throughput method to screen for fungal endoglucanase activity in E. coli. The method relies on a simple visual readout of substrate degradation, does not require enzyme purification, and is highly scalable. This allows for the rapid screening of large libraries of enzyme variants.
