Limb Bud Grafting

Limb bud grafting is an embryological transplantation technique in which a developing limb bud is moved between embryos to test how cells form and pattern the limbs. The procedure relies on microsurgical removal and placement of donor tissue into a host embryo, allowing researchers to observe interactions between transplanted cells and surrounding signals during development. In genetics and developmental biology, grafts can reveal whether tissue retains a predetermined fate or responds to local positional cues, including signaling from organizing regions and neighboring tissues. These experiments have helped clarify cell fate, tissue specification, growth regulation, and the genetic mechanisms that shape vertebrate limb development.

By grafting beads soaked in growth factors or specific inhibitors of signaling pathways into developing embryos it is possible to directly test their effects in vivo. In this protocol beads are grafted into the limb bud to determine the effects of these molecules on gene expression and signal transduction.
