Chimeric Mouse Generation

Chimeric mouse generation is a research method that produces mice containing cells or tissues from genetically distinct sources, enabling investigators to study immune function in a controlled living system. In a common approach, hematopoietic stem and progenitor cells are introduced into an immunodeficient recipient, where they engraft, proliferate, and reconstitute immune cell populations; other models use embryonic cell contribution or tissue transplantation. These mice help distinguish cell-intrinsic from environmental effects in immunology and provide platforms for investigating pathogen infection, host immune responses, immune development, and therapeutic strategies. Their design can also model human immune components when appropriately sourced cells are used.

We describe a protocol whereby busulfan conditioning permits the bone marrow of a recipient mouse to be replaced with bone marrow cells from donor mice ubiquitously expressing green fluorescent protein, in the absence of irradiation. This technique is useful to study bone marrow cell accumulation in the central nervous system.

An Ex Vivo Technique to Generate Tumor-Specific Chimeric Antigen Receptor T Cells

2025

This video demonstrates a technique for the ex vivo generation of tumor antigen-specific chimeric antigen receptor (CAR) T cells. Retroviral vectors carrying transgenic RNA are mixed with spleen cells in a medium supplemented with interleukin-2 to promote T-cell survival and proliferation. The mixture is then transferred to a plate coated with recombinant human fibronectin fragments and centrifuged to facilitate the fusion of the virus with the cell, allowing the RNA to integrate into the host...
