Dendritic Cell Analysis

Dendritic cell analysis is the study of dendritic cell identity, phenotype, function, and interactions within the immune system, making it essential for understanding how innate and adaptive immunity are linked. Researchers typically isolate cells from tissues or cultures and assess surface markers, antigen uptake, maturation, cytokine production, and antigen presentation using methods such as flow cytometry, microscopy, and functional assays. In immunology and infection research, these analyses reveal how pathogens influence dendritic cell activation and T-cell priming, helping clarify host defense, immune evasion, inflammation, and vaccine responses. The Analysis of Purkinje Cell Dendritic Morphology in Organotypic Slice Cultures

We present a protocol that permits to view and to quantitatively asses the morphology of the dendritic tree of individual Purkinje cells grown in organotypic cerebellar slice cultures. This protocol is intended to promote studies on the mechanisms of Purkinje cell dendritic development.

We describe a strategy to monitor maturation and migration of pulmonary dendritic cells in response to ovalbumin in the setting of ovalbumin induced allergic airway inflammation. This strategy can be modified to assess migration of pulmonary dendritic cells in settings of infection.

This video demonstrates a protocol for assessing the activation of dendritic cells using immunoglobulin G (IgG)-coated tumor cells. Dendritic cells internalize the IgG-coated tumor cells, process the tumor antigens, and present them on the surface through MHC-II molecules, along with the co-expression of the costimulatory molecule CD86. These dendritic cells then undergo MHC-II and CD86-targeted immunostaining and flow cytometry analysis to identify their activation state.

Here a protocol is presented to measure bacterial capture by CD4+ T cells which occurs during antigen presentation via transinfection from pre-infected dendritic cells (DC). We show how to perform the necessary steps: isolation of primary cells, infection of DC, DC/T cell conjugate formation, and measurement of bacterial T cell transfection.
