Flow Cytometry Sorting

Flow cytometry sorting is a cell-separation technique that identifies and isolates individual cells according to physical properties and fluorescent markers, enabling precise analysis of heterogeneous populations. In a flow cytometer, cells pass single file through laser beams after hydrodynamic focusing; detectors measure light scatter and fluorescence, while selected cells are enclosed in droplets that receive an electrical charge and are deflected into collection vessels by an electrostatic field. In immunology and infection research, this process enriches defined immune-cell subsets, separates infected from uninfected cells, and supports studies of antigen expression, host-pathogen interactions, immune activation, and cellular function. Sorted populations can then undergo culture, molecular profiling, or functional assays.

Step-specific Sorting of Mouse Spermatids by Flow Cytometry

We describe a sorting strategy for mouse spermatids using flow cytometry. Spermatids are sorted into four highly pure populations, including round (spermiogenesis steps 1-9), early elongating (spermiogenesis steps 10-12), late elongating (spermiogenesis steps 13-14) and elongated spermatids (spermiogenesis steps 15-16). DNA staining, size and granulosity are used as selection parameters.

In this video, we describe a protocol for the flow cytometry-based sorting of naïve B cell, memory B cell, and plasmablast plasma cell subpopulations from human peripheral blood B cells. The different B cell subpopulations can be distinguished based on fluorophore-based antibody binding to the CD19, CD27, and CD38 receptors expressed on the B cell surfaces, followed by cell sorting.

An efficient method to obtain highly purified viable meiotic fractions from mouse testis is described, which combines a refined cell dissociation protocol with fluorescent activated cell sorting (FACS). This method takes advantage of differences in the DNA content and nuclear density of discrete meiotic fractions.

Automation of the Micronucleus Assay Using Imaging Flow Cytometry and Artificial Intelligence

The micronucleus (MN) assay is a well-established test for quantifying DNA damage. However, scoring the assay using conventional techniques such as manual microscopy or feature-based image analysis is laborious and challenging. This paper describes the methodology to develop an artificial intelligence model to score the MN assay using imaging flow cytometry data.
