Lung Immune Cells

Lung immune cells are specialized leukocytes that protect the respiratory system from inhaled pathogens, particles, and tissue damage while helping maintain airway function. They include alveolar macrophages, dendritic cells, lymphocytes, and other immune populations that recognize danger signals, engulf microbes, present antigens, and coordinate inflammation through cytokine signaling. Their responses must balance rapid antimicrobial defense with regulation that limits injury to delicate gas-exchange surfaces. Studying lung immune cells is central to immunology and infection research, supporting investigation of pneumonia, tuberculosis, viral respiratory disease, asthma, and chronic lung inflammation, as well as the development of diagnostic tools and targeted therapies.

An In Vitro Method to Identify Innate and Adaptive Immune Cells Residing in Murine Lungs

This video demonstrates an in vitro technique to identify immune cells residing in murine lungs. After isolating target immune cells from murine lung tissue, they are surface-labeled using a cocktail of fluorophore-tagged antibodies against specific markers on innate and adaptive immune cells. Additionally, monocytes, macrophages, and dendritic cells are labeled using an intracellular marker. The various immune cell types are then identified using flow cytometry.

Isolation of Functional Cardiac Immune Cells

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.

We describe a method for generating Precision-cut Lung Slices (PCLS) and immunostaining them to visualize the localization of various immune cell types in the lung. Our protocol can be extended to visualize the location and function of many different cell types under a variety of conditions.
