Tpck Trypsin

TPCK trypsin is a purified serine protease preparation treated with tosyl phenylalanyl chloromethyl ketone (TPCK) to remove contaminating chymotrypsin-like activity while retaining trypsin function. Trypsin hydrolyzes peptide bonds on the carboxyl side of lysine and arginine residues, and TPCK treatment selectively inactivates chymotrypsin through irreversible modification, improving enzymatic specificity. In immunology and infection research, TPCK trypsin is used to process proteins, dissociate cells, and activate viral glycoproteins, including influenza hemagglutinin, during cell-culture experiments. Controlled proteolysis supports studies of viral entry, replication, antigen processing, and host-pathogen interactions while reducing confounding effects from unwanted proteases.
