Intestinal Graft

An intestinal graft is transplanted intestinal tissue used to restore digestive and absorptive function when a patient’s own intestine cannot support adequate nutrition, making it an important treatment in severe intestinal failure. During transplantation, surgeons connect the graft’s blood vessels and intestinal tract to the recipient, while immunosuppressive therapy limits immune recognition of donor antigens and reduces rejection. Intestinal grafts can enable enteral feeding, reduce dependence on parenteral nutrition, and improve quality of life, but they require careful monitoring for rejection, infection, and graft dysfunction. Research continues to improve donor selection, immune management, and long-term graft survival.

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.

In this video protocol we give a step by step explanation of lentiviral transduction in organoids of primary intestinal epithelium and of processing and downstream analysis of these cultures by quantitative RT-PCR, RNA-microarray and immunohistochemistry.

Here, we present a protocol that describes allogeneic hematopoietic stem cell transplantation and allows repetitive mini-endoscopic evaluations of the distal colon in situ for the presence, characteristics, and severity of colonic inflammation within live mice suffering from intestinal graft-versus-host disease.

To improve our knowledge of cellular and molecular neotissue formation, a murine model of the TEVG was recently developed. The grafts were implanted as infrarenal vena cava interposition grafts in C57BL/6 mice. This model achieves similar results to those achieved in our clinical investigation, but over a far shortened time-course.
