Rgd Peptide Inhibition

RGD peptide inhibition is a strategy that uses peptides containing the arginine-glycine-aspartic acid sequence to interfere with integrin-mediated cell interactions, which regulate adhesion, migration, and signaling. RGD peptides compete with extracellular matrix proteins such as fibronectin and vitronectin for binding to integrins, occupying their recognition sites and reducing downstream cellular responses. In medicine, this approach helps investigate integrin function and supports the development of therapies that target tumor angiogenesis, thrombosis, inflammation, and abnormal tissue remodeling. It also guides the design of targeted drug-delivery systems and biomaterials that control cell attachment.

The αvβ3 integrin is a type of adhesion protein that is highly expressed on activated endothelial cells undergoing angiogenesis. Thus, evaluating the integrity of the integrin is of great interest in oncology. Here, we introduce a method to prepare 68Ga-labeled radiopeptides and a method to assess its biological effectiveness.

We present a protocol for the synthesis of RGD-functionalized hydrogels as devices for cell and drug delivery. The procedure involves copper catalyzed alkyne-azide cycloaddition (CuAAC) between alkyne-modified polyacrylic acid (PAA) and a RGD-azide derivative. The hydrogels are formed using microwave-assisted polycondensation and their physicochemical properties are investigated.
