Electrode Track Tracing

Electrode track tracing is a neuroanatomical method used to locate and document the path and final position of an electrode in brain tissue, an essential step for interpreting neural recordings or stimulation results. After an experiment, researchers mark the electrode tip or track, often by passing a small electrical current to create a localized lesion, then fix, section, and stain the tissue for microscopic examination. Comparing the trace with anatomical landmarks confirms the targeted region and helps relate physiological effects to specific neural circuits. This verification supports accurate data interpretation, improves experimental reproducibility, and strengthens studies of brain function and connectivity.

We describe a technique to label neurons and their processes via anterograde or retrograde tracer injections into brain nuclei using an in vitro preparation. We modified an existing method of in vitro tracer electroporation by taking advantage of fluorescently labeled mouse mutants and basic optical equipment in order to increase labeling accuracy.

How neuronal networks are established in the embryonic brain is a fundamental question in developmental neurobiology. Here we combined an electroporation technique with novel genetic tools, such as Cre/Lox–plasmids and PiggyBac-mediated DNA transposition system in the avian hindbrain to label dorsal interneurons and track their axonal projections and synaptic targets at various developmental stages.
