Stereotaxic Syringe Pump

Stereotaxic syringe pumps are precision instruments that deliver minute, controlled fluid volumes to defined anatomical locations, making them important for reproducible experimental manipulations in neuroscience. They combine a motor-driven syringe with a stereotaxic apparatus: the frame positions a needle using three-dimensional coordinates, while calibrated plunger movement controls infusion rate and volume, often at very low flow rates that limit tissue disruption. Researchers use them to administer drugs, viral vectors, tracers, or other solutions into brain regions for studies of neural circuits, behavior, disease mechanisms, and therapeutic strategies. Accurate targeting and consistent delivery improve experimental comparability while reducing off-target effects.

Here we present a protocol to construct a pressure-controlled syringe pump to be used in microfluidic applications. This syringe pump is made from an additively manufactured body, off-the-shelf hardware, and open-source electronics. The resulting system is low-cost, straightforward to build, and delivers well-regulated fluid flow to enable rapid microfluidic research.

The monitoring of extracellular neurotransmitter levels in distinct brain regions of freely moving animals offers insights on the link between neurotransmitter release and behavior. In vivo microdialysis coupled with electrochemical detection provides excellent anatomical and chemical resolution; and information on how basal neurotransmission is altered by pharmacological or physiological manipulations.
