Synthetic Libraries

Synthetic libraries are deliberately designed collections of diverse biomolecules or chemical compounds used to identify candidates with useful properties. In immunology and infection research, synthetic antibody libraries generate sequence diversity by varying antigen-binding regions, which are displayed on platforms such as phage or yeast and selected for binding to a target antigen. Repeated rounds of binding, washing, and amplification enrich clones with desirable specificity and affinity. This approach supports the rapid isolation of antibodies against pathogens, toxins, or immune molecules, enabling diagnostic development, pathogen characterization, therapeutic discovery, and the engineering of reagents with properties that may be difficult to obtain from natural immune repertoires.

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
