Bleomycin Induction

Bleomycin induction is a research method that uses the chemotherapeutic agent bleomycin to trigger tissue injury and model disease processes, most commonly pulmonary fibrosis. Bleomycin generates iron-dependent reactive oxygen species that cause DNA strand breaks, initiating inflammatory signaling, epithelial damage, fibroblast activation, and excessive collagen deposition. In medicine and biomedical research, this approach helps investigators study the progression of fibrotic disease, identify cellular and molecular mechanisms, and evaluate potential antifibrotic treatments. Its controlled, experimentally reproducible injury response makes it valuable for linking tissue damage with remodeling, although findings must be interpreted carefully because experimental models do not fully reproduce human disease.

Here we present an effective method to investigate the antifibrotic activity of intravenously infused human mesenchymal stromal cells obtained from the whole umbilical cord following the induction of lung injury by an endotracheal injection of bleomycin in C57BL/6 mice. This protocol can be easily extended to the preclinical testing of other therapeutics.
