Bacterial Display Library

Bacterial display libraries are collections of genetically engineered bacteria that present diverse peptides or proteins on their cell surfaces, enabling rapid selection of molecules with desired binding or functional properties. Typically, DNA encoding each candidate is fused to a gene for a bacterial surface anchor, so the encoded product is displayed while its nucleotide sequence remains inside the same cell, preserving a genotype–phenotype link. Researchers can expose the population to a target, wash away nonbinders, recover attached cells, and amplify their DNA through repeated selection cycles. This platform supports antibody engineering, enzyme optimization, ligand discovery, and studies of protein–protein interactions in biochemistry.

Bacterial Inner-membrane Display for Screening a Library of Antibody Fragments

We provide a method to simultaneously screen a library of antibody fragments for binding affinity and cytoplasmic solubility by using the Escherichia coli twin-arginine translocation pathway, which has an inherent quality control mechanism for intracellular protein folding, to display the antibody fragments on the inner membrane.

Semi-automated Biopanning of Bacterial Display Libraries for Peptide Affinity Reagent Discovery and Analysis of Resulting Isolates

Biopanning bacterial display libraries is a proven technique for discovery of peptide affinity reagents, a robust alternative to antibodies. The semi-automated sorting method herein has streamlined biopanning to decrease the occurrence of false positives. Here we illustrate the thought process and techniques applied in evaluating candidates and minimizing downstream analysis.

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.
