Bleomycin Preconditioning

Bleomycin preconditioning is a research method that uses prior exposure to bleomycin to establish a controlled DNA-damage or cellular-stress state before a subsequent experiment or treatment. Bleomycin binds DNA and, in the presence of metal ions and oxygen, generates reactive oxygen species that produce single- and double-strand breaks, activating checkpoint, repair, senescence, or apoptosis pathways. In cancer research, this approach helps standardize cellular stress when evaluating anticancer agents, studying treatment resistance, or examining how tumors and surrounding tissues respond to additional challenges. Dose and timing influence whether preconditioning produces transient stress, cell death, or persistent changes in cell behavior.

We seek to define the neural immune signaling responsible for cold-preconditioning as means to identify novel targets for therapeutics development to protect brain before injury onset. We present strategies for such work that require biological systems, experimental manipulations plus technical capacities that are highly reproducible and sensitive.

Marrow stromal cells (MSCs) with neural potential exist within the bone marrow. Our protocol enriches this population of cells via hypoxic preconditioning and thereafter directs them to become mature Schwann cells.
