This manuscript presents a detailed protocol for the fabrication of an emerging three-dimensional hepatocyte culture platform, the inverted colloidal crystal scaffold, and the concomitant techniques to assess hepatocyte behavior. The size-controllable pores, interconnectivity and ability to conjugate extracellular matrix proteins to the poly(ethylene glycol) (PEG) scaffold enhance Huh-7.5 cell performance.
Het vermogen om hepatocytfunctie handhaven in vitro, met het oog op het testen van cytotoxiciteit xenobiotica, bestuderen virusinfectie en ontwikkeling van geneesmiddelen gericht op de lever, is een platform waarin cellen een gedegen biochemische en mechanische signalen. Recente leverweefsel technische systemen zijn driedimensionale (3D) steigers samengesteld uit natuurlijke of synthetische hydrogels gebruikt, gezien hun hoge waterretentie en hun vermogen om de mechanische stimuli die nodig zijn door de cellen. Er is toenemende interesse in de omgekeerde colloïdale kristal (ICC) scaffold is een recent; hoge ruimtelijke organisatie homotypische en heterotypische cel interacties, en cel-extracellulaire matrix (ECM) interactie mogelijk maakt. Hierin beschrijven we een protocol bij het ICC schavot te fabriceren met behulp van poly (ethyleen glycol) diacrylaat (PEGDA) en het deeltje uitspoeling methode. In het kort wordt een rooster gemaakt van microsfeer deeltjes, waarna een pre-polymer wordt toegevoegd, goed gepolymeriseerd, en de deeltjes worden vervolgens verwijderd of uitgeloogd, onder toepassing van een organisch oplosmiddel (bijvoorbeeld tetrahydrofuran). De ontbinding van het rooster resulteert in een zeer poreus schavot met gecontroleerde poriën en interconnectivities waarmee media om cellen gemakkelijker te bereiken. Deze unieke structuur stelt hoog specifiek oppervlak voor de cellen te hechten aan en eenvoudige communicatie tussen poriën, en de mogelijkheid het bekleden van de PEGDA ICC scaffold eiwitten vertoont een duidelijk effect op celprestatie. We analyseren de morfologie van de steiger en de hepatocarcinoma cel (Huh-7,5) gedrag inzake levensvatbaarheid en functie om het effect van ICC structuur en ECM coatings verkennen. Kortom, dit document geeft een gedetailleerde protocol van een nieuwe steiger die brede toepassingen in tissue engineering, met name lever tissue engineering heeft.
De lever is een zeer gevasculariseerde orgel met een veelheid aan functies, ontwenning van het bloed, metabolisme van xenobiotica en de productie van serumeiwitten. Leverweefsel heeft een complexe driedimensionale (3D) microstructuur, bestaande uit meerdere celtypes, galcanaliculi, sinusoïden, en zones van verschillende samenstelling en biomatrix verschillende zuurstofconcentraties. Gezien deze gedetailleerde structuur is het moeilijk om een goede lever model in vitro 1 te creëren. Er is echter een toenemende vraag naar functionele in vitro modellen hosting menselijke hepatocyten als platform voor het testen geneesmiddelentoxiciteit 2 en studeren ziekten geassocieerd met lever 3.
Huidige leverweefsel techniek platforms de complexiteit van de lever vereenvoudigd door het isoleren van één of gericht op enkele van de lever parameters, namelijk co-celkweek 4, biochemische samenstelling van de zonal microenvironments 5, stroomdynamica 6,7 en de configuratie van de biomatrix 8. Configuratie van de biomatrix kan worden onderverdeeld in parameters zoals scaffold materialen, samenstelling van de extracellulaire matrix (ECM) proteïnen, matrix stijfheid en het ontwerp en constructie van de steiger. Er is een toename in tissue engineering studies met synthetische hydrogelen, met name poly (ethyleenglycol) (PEG) hydrogelen 9 is, krijgt de mogelijkheid om af te stemmen de hydrogel van mechanische eigenschappen, biologische en afbraaksnelheid. Ten aanzien van lever-gerelateerde onderzoek, werd het biocompatibele hydrogel aangevraagd virusinfectie studie van leverziekte 3. De omgekeerde kristal colloïdale (ICC) 14 steiger is een zeer georganiseerde 3D scaffold voor celcultuur die voor het eerst in de vroege jaren 2000 werd geïntroduceerd. unieke structuur van de steiger kan worden toegeschreven aan het eenvoudige fabricageproces onder toepassing van een colloïdaal kristal, een geordende rooster van colloïdale deeltjes met variabele diameter. In het kort, om het proces te vatten, deeltjes zijn netjes gerangschikt en gloeien met behulp van warmte om een rooster te vormen. De uitloging van dit rooster door een organisch oplosmiddel, in een gepolymeriseerde hydrogel leidt tot hexagonaal gepakte bolvormige holten 15 met een groot oppervlak. Deze sterk geordende scaffold is eerder gemaakt met zowel synthetische als natuurlijke materialen, inclusief maar niet beperkt tot, poly (acrylamide) 16-21, poly (melkzuur-co-glycolzuur) 15,22-30, Poly (ethyleenglycol) 31,32, poly (2-hydroxyethylmethacrylaat) 21,33-35, 36-39 en chitosan. ICC scaffolds van non-fouling materialen vaak cellulaire sferoïden binnen de holten 14,23,40 bevorderen. Meerdere celtypen is aangetoond succesvol prolifereren, differentiëren en functie binnen deze configuratie, waaronder chondrocyten 41, beenmergstromacellen 42 en stamcellen 43,44. Hepatocyt betrekking hebben studies uitgevoerd met ICC scaffolds uit Na 2 SiO 3 en poly (acrylamide), maar geen PEG. Met eenvoudige bioconjugatie strategieën (dat wil zeggen, amine koppeling door middel van EDC / NHS), kunnen ECM eiwitten geconjugeerd PEG-gebaseerde steigers worden vervaardigd, die kan blijken te zijn meer mobiele bindingsplaatsen naar een meer in vivo-achtige omgeving en de leverfunctie te verbeteren.
In dit manuscript en de bijbehorende video, we detail de fabricage van het ICC steigergebruik poly (ethyleenglycol) diacrylaat (PEGDA) hydrogel en een polystyreen microsfeer rooster, geoptimaliseerd voor hepatocarcinoma (Huh-7,5) cultuur. We tonen de verschillen tussen het algemeen klevend kale PEGDA ICC scaffolds en de met collageen beklede PEGDA ICC scaffold qua steiger topologie en celprestaties. Levensvatbaarheid van de cellen en functie zijn kwalitatief en kwantitatief gemeten om Huh-7.5 cel gedrag te beoordelen.
Scaffolds evolueren snel alle fysische en biochemische signalen nodig te regenereren, handhaven of te herstellen weefsel ter uitvoering van orgaanvervanging, bestuderen ziekte, de ontwikkeling van geneesmiddelen en vele anderen 57 verschaffen.
