Dit experiment wordt een anatomisch beperkt-Liquor (aMEG)-methode oscillerende dynamiek van de hersenen en lange-afstands functionele synchrony gedurende betrokkenheid van cognitieve controle te onderzoeken als een functie van acute alcohol gedronken.
Besluitvorming is gebaseerd op dynamische interacties van gedistribueerde, hoofdzakelijk frontale hersengebieden. Uitgebreide bewijs van functionele magnetische resonantie imaging (fMRI) studies geeft aan dat de voorste cingularis (ACC) en de laterale prefrontale cortices (latPFC) zijn essentiële knooppunten subserving cognitieve controle. Echter, vanwege de beperkte temporele resolutie, fMRI kan niet nauwkeurig overeen met de timing en aard van hun vermoedelijke samenspel. De huidige studie combineert gedistribueerde bron modellering van het stoffelijk precieze Liquor (MEG) signaal met structurele MRI in de vorm van “hersenen films” aan: (1) raming de corticale gebieden bij cognitieve controle betrokken (“waar”), (2) karakteriseren hun temporele sequentie (“wanneer”), en (3) de oscillerende dynamiek van hun Neurale interacties in real-time te kwantificeren. Stroop interferentie werd geassocieerd met grotere gebeurtenis-gerelateerde theta (4-7 Hz) macht in de ACC tijdens conflictdetectie gevolgd door aanhoudende gevoeligheid naar cognitieve eisen in de ACC en latPFC tijdens de voorbereiding van de integratie en reactie. Een analyse van fase-vergrendeling bleek co-oscillatory interacties tussen deze gebieden met vermelding van hun toegenomen neurale synchrony in theta-band tijdens conflict-inducerende onlogisch proeven. Deze resultaten bevestigen dat theta oscillaties lange-afstands synchronisatie, die nodig zijn voor de integratie van bovenaf invloeden tijdens cognitieve controle van fundamenteel belang zijn. MEG weerspiegelt neurale activiteit direct, waardoor het geschikt is voor farmacologische manipulaties in tegenstelling tot de fMRI die gevoelig is voor vasoactieve kunstdiscours. In de huidige studie, werden gezonde sociale drinkers gegeven een matige alcohol dosis en placebo in een binnen-onderwerp-ontwerp. Acute intoxicatie verzwakt theta macht om Stroop conflict en dysregulated co oscillaties tussen de ACC en latPFC, waarin wordt bevestigd dat alcohol schadelijk is voor neurale synchrony subserving cognitieve controle. Het interfereert met doelgericht gedrag dat leiden gebrekkige zelfbeheersing tot kan, bij te dragen aan het drinken van dwangmatig. Kortom, deze methode kan inzicht geven in real-time interacties tijdens de cognitieve verwerking en de selectieve gevoeligheid voor farmacologische uitdaging over relevante neurale netwerken kunnen typeren.
Het algemene doel van deze studie is het effect van acute alcohol gedronken op spatio-temporele veranderingen in de hersenen oscillerende dynamiek en lange-afstands functionele integratie tijdens cognitieve controle te onderzoeken. De werknemer multimodale aanpak imaging combineert Liquor (MEG) en structurele magnetische resonantie beeldvorming (MRI) inzicht te geven in de neurale basis van besluitvorming met hoge temporele precisie en op het niveau van een interactief systeem.
Flexibel gedrag maakt het mogelijk aan te passen aan de veranderende eisen van de contextuele en strategisch schakelen tussen verschillende taken en eisen in overleg met iemands oogmerken en doelstellingen. De capaciteit om te onderdrukken automatische reacties ten gunste van doel-relevante maar niet-gewone acties is een wezenlijk aspect van cognitieve controle. Uitgebreide aanwijzingen dat het subserved is door een overwegend frontale corticale netwerk, met de voorste cingularis cortex (ACC) als een centraal knooppunt in deze interactieve netwerk1,2,3,4. Terwijl de overvloedige anatomische connectiviteit tussen de ACC en laterale frontale cortices goed beschreven5,6, de functionele kenmerken van de communicatie tussen deze regio’s tijdens cognitieve controle, reactie selectie is en uitvoering, zijn slecht begrepen.
Het zeer invloedrijke conflict controle theorie7,8 stelt dat cognitieve controle vloeit uit een dynamische interactie tussen de mediale en laterale prefrontale cortices voort. Deze account pretendeert dat de ACC conflict tussen de concurrerende vertegenwoordigingen controleert en houdt zich bezig de laterale prefrontale cortex (latPFC) uitvoering van de controle van de reactie te optimaliseren. Deze account is echter voornamelijk gebaseerd op de functionele MRI (fMRI) studies met behulp van het bloed oxygenatie niveau afhankelijk (BOLD) signaal. Het signaal van fMRI-vet is een uitstekende ruimtelijke mapping tool, maar de temporele resolutie is beperkt omdat het getuigt van regionale hemodynamische wijzigingen gemedieerd door neurovasculaire koppeling. Dientengevolge, ontvouwen de vet signaal wijzigingen op een veel langzamere tijdschaal (in seconden) dan de onderliggende neurale gebeurtenissen (in milliseconden)9. Bovendien, het vet signaal is gevoelig voor alcohol van vasoactieve effecten10 en kan vertegenwoordigen niet nauwkeurig de omvang van de neurale veranderingen, waardoor het minder geschikt voor studies van acute alcohol gedronken. Daarom is het vermoedelijke samenspel tussen de mediale en laterale prefrontale cortices en de gevoeligheid voor alcohol intoxication moeten worden onderzocht volgens methoden die neurale gebeurtenissen in een stoffelijk nauwkeurige manier registreren. MEG heeft een uitstekende temporele resolutie omdat hierin direct postsynaptisch stromingen. De anatomisch beperkt-MEG (aMEG) methodiek hier werkzaam is een multimodale benadering die combineert verdeeld bron modellering van het signaal van de MEG met structurele MRI. Het staat voor de schatting van waar die de conflict – en drank-gerelateerde hersenen oscillerende wijzigingen zich voordoen en om te begrijpen van de temporele sequentie (“wanneer”) van de betrokken neurale componenten.
Besluitvorming is afhankelijk van de interacties van gedistribueerde hersengebieden die dynamisch om te gaan met hogere eisen op cognitieve controle betrokken zijn. Unidirectioneel om te schatten gebeurtenis-gerelateerde veranderingen in lange-afstands synchronie tussen twee corticale gebieden is het berekenen van hun fase als een index van hun co oscillaties11,12te koppelen. De huidige studie toegepast een fase-vergrendeling analyse om te testen het basisprincipe van het conflict controle theorie door het onderzoek van de co-oscillatory interactie tussen de ACC en latPFC. Neurale oscillaties in theta bereik (4-7 Hz) zijn gekoppeld aan cognitieve controle en voorgesteld als een fundamentele mechanisme ter ondersteuning van de lange-afstands synchronisatie, die nodig zijn voor de top-down cognitieve verwerking13,14, 15,16. Ze worden gegenereerd in de prefrontale gebieden als functie van de moeilijkheidsgraad van de taak en zijn aanzienlijk verzwakt door acute alcohol intoxication17,18,19,20.
Langdurig overmatig alcoholgebruik is gekoppeld aan een scala van cognitieve tekorten met prefrontale circuits wordt vooral getroffen21,22. Acute alcohol gedronken is schadelijk voor cognitieve controle onder voorwaarden van verhoogde moeilijkheidsgraad, ambiguïteit, of die reactie onverenigbaarheid17,23,24 induceren. Door het beïnvloeden van besluitvorming, alcohol kan interfereren met doelgericht gedrag, kan resulteren in slechte zelfbeheersing en meer drinken, en kan ook bijdragen aan verkeer – of werk-gerelateerde risico’s25,26,27 . De huidige studie maakt gebruik van een aMEG aanpak voor het meten van de oscillerende activiteit in theta-band en synchronie tussen de belangrijkste uitvoerende gebieden met uitstekende temporele resolutie. De effecten van alcohol op theta activiteit en co oscillaties tussen de ACC en de latPFC worden onderzocht als een functie van conflict ontlokte door de Stroop interferentie taak. We veronderstellen dat hogere cognitieve eisen geassocieerd met meer functionele synchrony worden en dat alcohol-geïnduceerde disregulatie van synchrone activiteit van de mediale en laterale prefrontale cortices ten grondslag ligt aan afboekingen in cognitieve controle.
De multimodale imaging methode die in deze studie wordt gebruikt omvat gedistribueerde bron modellering van het stoffelijk precieze MEG signaal samen met ruimtelijke beperkingen van inverse schattingen afgeleid van elke deelnemer structurele MRI. De aanpak van de aMEG combineert de sterke punten van deze technieken bieden inzicht in de spatio-temporele stadia van oscillerende dynamiek en de lange-afstands integratie subserving cognitieve controle.
