Cette expérience utilise une méthode de magnétoencéphalographie anatomiquement sont limitées (aMEG) d’examiner la dynamique oscillatoire de cerveau et de la synchronie fonctionnelle à long terme au cours de l’engagement du contrôle cognitif en fonction d’une intoxication alcoolique aiguë.
Prise de décision s’appuie sur les interactions dynamiques de régions distribuées, essentiellement frontal du cerveau. Nombreux éléments de preuve des études d’imagerie de résonance magnétique fonctionnelle (IRMf) indique que le cingulaire antérieur (ACC) et le cortex préfrontal latéral (latPFC) des nœuds essentiels favorisant le contrôle cognitif. Toutefois, en raison de sa résolution temporelle limitée, IRMf ne peut pas refléter le calendrier et la nature de leur interaction présumée. La présente étude combine source distribué modélisation du signal temporel précis magnétoencéphalographie (MEG) avec IRM structurelle sous forme de « films de cerveau » à : (1) estimation les aires corticales impliquées dans le contrôle cognitif (« où »), (2) caractérisent leur séquence temporelle (« quand ») et (3) quantifier la dynamique oscillatoire de leurs interactions neurales en temps réel. Interférence de Stroop a été associée à une plus grande puissance liées à l’événement thêta (4-7 Hz) de l’ACC pendant la détection de conflit suivie de sensibilité soutenue aux exigences cognitives dans l’ACC et latPFC lors de la préparation de l’intégration et de la réponse. Une analyse de verrouillage de phase a révélé des interactions co-oscillatory entre ces zones indiquant leur synchronie neuronale accrue dans la bande thêta pendant les essais incongrus inductrices de conflit. Ces résultats confirment que les oscillations thêta sont fondamentales pour la synchronisation à long terme nécessaire pour intégrer des influences de haut en bas au cours du contrôle cognitif. MEG reflète l’activité neurale directement, ce qui le rend adapté aux manipulations pharmacologiques contrairement aux IRMf qui est sensible aux vasoactifs confond. Dans la présente étude, des buveurs sociaux sains ont reçu une dose modérée d’alcool et le placebo dans une conception intra-sujet. Intoxication aiguë atténuée thêta pouvoir Stroop conflit et dysrégulation co oscillations entre le CAC et le latPFC, confirmant que l’alcool est nuisible à la synchronie neuronale favorisant le contrôle cognitif. Il interfère avec le comportement ciblé qui peut conduire à la maîtrise de soi déficiente, contribuant à une consommation compulsive. En somme, cette méthode peut fournir la perspicacité dans les interactions en temps réel au cours du traitement cognitif et peut caractériser la sensibilité sélective au défi pharmacologique à travers les réseaux de neurones.
L’objectif général de cette étude est d’examiner les effets d’une intoxication alcoolique aiguë sur changements spatio-temporels dans la dynamique oscillatoire de cerveau et l’intégration fonctionnelle à long terme au cours du contrôle cognitif. Le travailleur multimodal d’imagerie approche combine magnétoencéphalographie (MEG) et structurels d’imagerie par résonance magnétique (IRM) pour donner un aperçu de la base neurale de prise de décision avec une grande précision temporelle et au niveau d’un système interactif.
Comportement flexible permet de s’adapter à l’évolution des exigences contextuelles et de basculer stratégiquement entre différentes tâches et exigences en accord avec ses intentions et objectifs. La capacité de supprimer les réponses automatiques en faveur des actions objectif pertinent mais non habituelle est un aspect essentiel du contrôle cognitif. Une preuve abondante donne à penser qu’il est effectué par un réseau cortical prédominance frontal, avec le cortex cingulaire antérieur (CCA) comme un point nodal dans ce réseau interactif1,2,3,4. Alors que la connectivité anatomique abondante entre le CAC et le cortex frontal latéral est bien décrit5,6, les caractéristiques fonctionnelles de la communication entre ces régions au cours du contrôle cognitif, la sélection de la réponse et exécution, sont mal compris.
Le conflit très influent théorie7,8 de surveillance propose que contrôle cognitif résulte d’une interaction dynamique entre le cortex préfrontal médial et latéral. Ce compte prétend que l’ACC surveille le conflit entre les représentations concurrentes et engage le cortex préfrontal latéral (latPFC) à mettre en œuvre le contrôle de la réponse et optimiser les performances. Toutefois, ce compte repose principalement sur les études de MRI (IRMf) fonctionnelles en utilisant le signal de niveau (BOLD) dépendant sang oxygénation. Le signal IRMf-BOLD est un outil de cartographie spatiale excellent, mais sa résolution temporelle est limitée parce qu’il reflète les modifications hémodynamiques régionale médiées par un couplage neurovasculaire. Par conséquent, les modifications de signal “BOLD” se déroulent sur une beaucoup plus lente échelle de temps (en secondes) que le sous-jacent des événements neuronaux (en millisecondes)9. En outre, le signal “BOLD” est sensible à effets vasoactifs10 de l’alcool et ne représente pas exactement l’ampleur des changements neurones, qui le rend moins approprié pour l’étude de l’intoxication alcoolique aiguë. Par conséquent, l’interaction présumée entre le cortex préfrontal médial et latéral et sa sensibilité à l’alcool intoxication besoin d’être examinée par des méthodes qui enregistrent les événements neurones de manière précise dans le temps. MEG a une excellente résolution temporelle, puisqu’elle reflète directement les courants postsynaptiques. La méthodologie de MEG (aMEG) contraint anatomiquement employée ici est une approche multimodale qui combine distribué de modélisation de la source du signal MEG avec IRM structurelle. Elle permet l’estimation où que les boisson-liées au conflit et cerveau oscillatoire changements se produisent et de comprendre la séquence temporelle (« quand ») des éléments neurones impliqués.
Prise de décision s’appuie sur les interactions entre les régions du cerveau distribués qui sont dynamiquement engagées pour faire face à des exigences accrues sur contrôle cognitif. Estimer les changements liés à l’événement en synchronie à longue distance entre les deux régions corticales consiste à calculer leur phase de couplage comme indice de leurs oscillations co11,12. La présente étude appliqué une analyse à verrouillage de phase pour tester le principe fondamental du conflit suivi théorie en examinant les interactions co-oscillatory entre le CAC et le latPFC. Oscillations neurales dans gamme thêta (4-7 Hz) sont associées à contrôle cognitif et ont été proposées comme un mécanisme fondamental soutenant la synchronisation à distance nécessaire pour un traitement cognitif vertical13,14, 15,16. Ils sont générés dans les zones préfrontal en fonction de la difficulté de la tâche et sont considérablement atténués par aiguë d’alcool intoxication17,18,19,20.
Consommation d’alcool excessive à long terme est associée à une variété de déficits cognitifs avec circuit préfrontal étant particulièrement touchées21,22. Une intoxication alcoolique aiguë est préjudiciable au contrôle cognitif dans les conditions de difficulté accrue, ambiguïté ou ceux qui induisent la réponse incompatibilité17,23,24. En affectant la prise de décisions, l’alcool peut-être interférer avec le comportement ciblé, peut entraîner mauvaise maîtrise de soi et la consommation accrue d’alcool et peut aussi contribuer au trafic – ou liées au travail risques25,26,27 . La présente étude utilise une approche aMEG pour mesurer l’activité oscillatoire en bande thêta et synchronisme entre les principaux domaines exécutifs avec une excellente résolution temporelle. Les effets de l’alcool sur l’activité thêta et co oscillations entre le CAC et le latPFC sont examinés en fonction du conflit suscité par la tâche d’interférence de Stroop. Nous émettons l’hypothèse qu’accrue des exigences cognitives sont associées à une plus grande synchronie fonctionnelle et ce dérèglement induite par l’alcool de l’activité synchrone de la cortex préfrontal médial et latéral sous-tend des déficiences dans le contrôle cognitif.
Le multimodal méthode utilisée dans cette étude d’imagerie comprend des sources modélisation du signal temporel précis MEG ainsi que les contraintes spatiales d’inverses estimations tirées de MRI structurelle de chaque participant. L’approche de l’aMEG combine les points forts de ces techniques pour donner un aperçu des étapes spatio-temporelle de la dynamique oscillatoire et l’intégration à long terme favorisant le contrôle cognitif.
