Nematode Communication

Nematode communication is the exchange of information among roundworms through chemical signals, physical cues, and environmental changes, enabling coordinated behavior and adaptation. Many nematodes release and detect pheromones called ascarosides, which can alter development, aggregation, dispersal, mating, and responses to stress through sensory neurons and downstream neural pathways. Studying these signaling systems in behavioral biology reveals how simple nervous systems generate social behaviors and make decisions in changing environments. This research also supports investigations of host-parasite interactions, population dynamics, and potential strategies for controlling agriculturally important or disease-causing nematodes.

Visualizing Bacteria in Nematodes using Fluorescent Microscopy

To study the mutualism between Xenorhabdus bacteria and Steinernema nematodes, methods were developed to monitor bacterial presence and location within nematodes. The experimental approach, which can be applied to other systems, entails engineering bacteria to express the green fluorescent protein and visualizing, using fluorescence microscopy bacteria within the transparent nematode.

Assaying Predatory Feeding Behaviors in Pristionchus and Other Nematodes

Behavioral assays provide powerful tools for understanding neuronal function. Here we present several protocols for quantifying predatory feeding behavior found in the model nematode Pristionchus pacificus and its relatives. Additionally, we provide methods for analyzing predatory feeding adaptations including mouth structures and teeth.

An In Vivo Assay to Detect Mitophagy in Transgenic Nematodes

This video demonstrates an in vivo assay to study mitophagy in Caenorhabditis elegans. Upon exposing the nematodes to a mitophagy-inducing drug, the occurrence of mitophagy is assessed by co-localization between DCT-1, a mitophagy receptor on the damaged mitochondria, and the autophagosomal membrane protein LGG-1.
