3d Cellular Assembly

3D cellular assembly is the organization of living cells into three-dimensional structures that more closely reproduce tissue architecture than conventional two-dimensional cultures. It works through coordinated cell-cell adhesion, extracellular matrix interactions, and biochemical or mechanical cues that guide cells to aggregate, self-organize, and maintain spatial relationships. In biochemistry, these assemblies provide models for studying signaling, differentiation, metabolism, and cell-matrix communication under physiologically relevant conditions. Applications include constructing spheroids, organoid-like tissues, and engineered tissue models for disease research, drug evaluation, and regenerative medicine, while improving analysis of how molecular processes change in a three-dimensional environment.

Micropatterning and Assembly of 3D Microvessels

This manuscript presents an injection molding method to engineer microvessels that recapitulate physiological properties of endothelium. The microfluidic-based process creates patent 3D vascular networks with tailorable conditions, such as flow, cellular composition, geometry, and biochemical gradients. The fabrication process and examples of potential applications are described.

We present protocols for the 3-dimensional (3D) encapsulation of cells within synthetic hydrogels. The encapsulation procedure is outlined for two commonly used methods of crosslinking (michael-type addition and light-initiated free radical mechanisms), as well as a number of techniques for assessing encapsulated cell behavior.

Construction of Modular Hydrogel Sheets for Micropatterned Macro-scaled 3D Cellular Architecture

We describe the fabrication of micropatterned hydrogel sheets using a simple process, which can be assembled and manipulated in a freestanding form. Using these modular hydrogel sheets, a simple macro-scaled 3D cell culture system can be generated with a controlled cellular microenvironment.

Engineering 3D Cellularized Collagen Gels for Vascular Tissue Regeneration

In this work, we present a technique for the rapid fabrication of living vascular tissues by direct culturing of collagen, smooth muscle cells and endothelial cells. In addition, a new protocol for the mechanical characterization of engineered vascular tissues is described.
