Urea Loading Dye

Urea loading dye is a sample-preparation reagent used in biochemistry to denature biomolecules and monitor their migration during gel electrophoresis. Its urea component disrupts hydrogen bonds and weakens noncovalent interactions, helping unfold proteins or nucleic acids, while tracking dyes indicate sample movement through the gel; density-enhancing agents help the sample settle into wells. By reducing structure-dependent differences, urea loading dye supports separation based primarily on molecular size in denaturing polyacrylamide or nucleic acid gels. It is therefore important for analyzing protein composition, assessing nucleic acid length, and preparing samples for reproducible electrophoretic comparisons.

Denaturing urea polyacrylamide gel electrophoresis is used to separate single-stranded DNA or RNA up to a limit of 500 nucleotides. Urea in combination with heat denatures samples and unstructured single strands migrate within the gel matrix according to their molecular weight.

Targeted-esterase induced dye loading (TED) supports the analysis of intracellular calcium store dynamics by fluorescence imaging. The method bases on targeting of a recombinant Carboxylesterase to the endoplasmic reticulum (ER), where it improves the local unmasking of synthetic low-affinity Ca2+ indicator dyes in the ER lumen.

In this video, Drosophila larvae nerve fibers are immersed in FM dye, which binds to cell membrane lipids. Suction microelectrode and electrical pulses stimulate the endocytosis process, incorporating the FM dye into newly formed endocytic vesicles. This method, which visualizes presynaptic stages of neurotransmission, is useful for studying synaptic function and neurotransmitter release.
