Anion Exchange

Anion exchange is the reversible replacement of one negatively charged ion, or anion, by another on a positively charged material or transport system. It occurs when electrostatic attraction temporarily retains anions and differences in concentration, charge balance, or chemical affinity drive their release and substitution; in biological membranes, specialized exchange proteins couple the movement of one anion to another. In biology, anion exchange supports electrolyte regulation, acid–base balance, and carbon dioxide transport, while anion-exchange chromatography separates proteins, nucleic acids, and other charged biomolecules according to binding strength. These principles help researchers analyze cellular transport and purify biological samples.

Source: Schaeffer, J. W., Chandler, et al. Detection of Viruses from Bioaerosols Using Anion Exchange Resin. J. Vis. Exp. (2018).This video demonstrates the detection of viruses from bioaerosols using an anion exchange resin. A liquid impinger containing the resin is used to capture viruses from the air. The resin is collected and treated with a virus lysis buffer containing carrier RNA. The lysate is transferred to a microcentrifuge tube for viral RNA isolation, followed by qRT-PCR to amplify...

Source: Laboratory of Dr. B. Jill Venton - University of Virginia Ion-exchange chromatography is a type of chromatography that separates analytes based on charge. A column is used that is filled with a charged stationary phase on a solid support, called an ion-exchange resin. Strong cation-exchange chromatography preferentially separates out cations by using a negatively-charged resin while strong anion-exchange chromatography preferentially selects out anions by using a positively-charged...

Purification of Extracellular Trypanosomes, Including African, from Blood by Anion-Exchangers (Diethylaminoethyl-cellulose Columns)

This method of trypanosome separation from blood depends on their surface charge being less negative than mammalian blood cells. Infected blood is placed and treated on an anion-exchanger column. This method, the most fitting diagnostic for African trypanosomiasis, provides purified parasites for immunological, biological, biochemical, pharmaceutical and molecular biology investigations.
