Babb Clearing Solution

Babb clearing solution, commonly written BABB, is an organic solvent mixture of benzyl alcohol and benzyl benzoate used to render biological specimens optically transparent for microscopy. It works by replacing water in fixed tissue with solvents that closely match the refractive index of cellular components, reducing light scattering and improving optical access through the sample. In biology, BABB clearing supports three-dimensional imaging of embryos, organs, and labeled tissues, often after fluorescent staining or genetic labeling, and helps reveal structures that are difficult to resolve in opaque specimens. Its use requires attention to solvent toxicity and possible effects on fluorescent signals.

Analysis of Coronary Vessels in Cleared Embryonic Hearts

We present a protocol for the analysis of coronary vessels in whole embryonic murine hearts up to E15.5, using standard immunological staining methods followed by optical clearance and confocal microscopy. This technique enables visualization of blood vessels throughout the entire heart without the need for time-consuming analysis of serial sections.

Multiphoton Microscopy of Cleared Mouse Brain Expressing YFP

Multiphoton microscopy of whole mouse organs is possible by optically clearing the organ before imaging, but not all protocols preserve the fluorescent signal of fluorescent proteins. Using an optical clearing method with ethanol-based dehydration and benzyl alcohol:benzyl benzoate clearing, we show high-resolution multiphoton images of whole mouse brain expressing YFP.

Imaging Cleared Intact Biological Systems at a Cellular Level by 3DISCO

To obtain high-resolution images of fluorescently labeled cells within large tissues, ideally, the biological samples should be imaged without sectioning. 3DISCO is a straightforward tissue clearing procedure based on sequential incubation with organic solvents. Upon clearing, the organs become transparent allowing an end-to-end laser scan of the specimen.

Imaging Cleared Embryonic and Postnatal Hearts at Single-cell Resolution

We describe a protocol to volumetrically image fluorescent protein labeled cells deep inside intact embryonic and postnatal hearts. Utilizing tissue-clearing methods in combination with whole mount staining, single fluorescent protein-labeled cells inside an embryonic or postnatal heart can be imaged clearly and accurately.
