Blood Immune Cells

Blood immune cells are leukocytes that circulate through the bloodstream and coordinate defense against pathogens, abnormal cells, and tissue injury. They include innate immune cells, such as neutrophils, monocytes, and natural killer cells, which respond rapidly through phagocytosis, inflammatory signaling, or target-cell destruction, as well as lymphocytes that recognize specific antigens and generate antibody- or cell-mediated adaptive responses. Studying these cells helps explain infection, inflammation, autoimmunity, and cancer, while blood-cell counts, flow cytometry, and functional assays support diagnosis, treatment monitoring, vaccine research, and the development of immunotherapies.

We describe a method combining immunomagnetic beads and fluorescence-activated cell sorting to isolate and analyze defined immune cell subpopulations of peripheral blood mononuclear cells (monocytes, CD4+ T cells, CD8+ T cells, B cells, and natural killer cells). Using this method, magnetic and fluorescently labeled cells can be purified and analyzed.

Isolating Cord Blood Hematopoietic Stem Cells to Generate Human Immune System Mice

This video demonstrates a technique for generating human immune system (HIS) mice. CD34+ hematopoietic stem cells (HSCs) are isolated from human cord blood buffy coat cells using antibody-coated magnetic beads. The CD34+ HSCs are expanded in a growth medium enriched with cytokines and cryopreserved. Finally, the thawed HSCs are injected into an immunodeficient mouse pup to establish the HIS mouse model.

A Flow Cytometry Assay to Identify Immune Cell Subsets in Peripheral Blood Mononuclear Cells

This video demonstrates a flow cytometry-based technique using two fluorochromes to identify CD4+ and CD8+ T cells, γδ T cells, B cells, natural killer (NK) cells, and monocytes in human peripheral blood mononuclear cells (PBMCs). Upon incubating the cells with fluorochrome-conjugated antibodies against the surface markers, the cell subsets are identified using an optimal flow cytometric gating strategy.

This method for isolating functional immune cells from the heart provides an alternative to the conventional methods of collagenase digestion, which causes unwanted immune cell activation, resulting in a decreased responsiveness of these cells. Our method of isolation yields functional cardiac immune cells by avoiding problems associated with enzymatic digestion.
