Bone Growth

Bone growth is the biological process by which bones increase in length, width, and strength, shaping the skeleton during development and maintaining its structure throughout life. Lengthwise growth occurs at epiphyseal growth plates, where cartilage cells proliferate, enlarge, and are replaced by mineralized bone; osteoblasts build new tissue, while osteoclasts resorb older bone during remodeling. This coordinated activity responds to genetic signals, hormones, mechanical loading, and nutrient availability, with growth-plate activity largely ending after puberty. Understanding bone growth supports research into skeletal development, fractures, osteoporosis, and growth disorders, while informing strategies for bone repair and regenerative medicine.

Here we present a surgical protocol in rabbits with the aim to assess bone substitution materials in terms of bone regeneration capacities. By using PEEK cylinders fixed onto rabbit skulls, osteoconduction, osteoinduction, osteogenesis and vasculogenesis induced by the materials may be evaluated either on live or euthanized animals.

Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
