Bone Visualization

Bone visualization is the imaging or labeling of skeletal tissue to reveal its structure, composition, and changes in biological systems. In radiography and computed tomography, mineralized matrix attenuates X-rays, producing contrast that maps bone density and architecture; microscopy and fluorescent stains can instead highlight cells, fibers, or newly deposited mineral. These approaches support studies of bone development, remodeling, fracture healing, and diseases such as osteoporosis by connecting tissue-level organization with cellular activity. Quantitative images can also provide measurements of thickness, volume, and mineral density, helping researchers evaluate skeletal health and assess experimental treatments.

Bioengineering of Humanized Bone Marrow Microenvironments in Mouse and Their Visualization by Live Imaging

A method to create and live-image different humanized bone-marrow niches in mice is presented. Based on the supportive niche created by human mesenchymal cells, the addition of human endothelial cells induces the formation of human vessels, while the addition of rhBMP-2 induces the formation of human-mouse chimeric mature bone tissue.

Animal models are frequently utilized to study cancer metastasis to bone. In this protocol we will describe two common methods of tumor inoculation for bone metastasis studies and briefly describe some of the analyses utilized to monitor and quantify these models.

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
