Cell-cell Communication

Cell-cell communication is the exchange of information between cells that coordinates activities such as growth, differentiation, movement, and responses to changing conditions. Cells communicate through direct contact, cell junctions, or signaling molecules released into the local environment or bloodstream; these signals bind specific receptors and trigger intracellular pathways that alter gene expression or cell behavior. In biology, this process helps organize tissues, regulate immune responses, and maintain homeostasis across multicellular organisms. Studying cell-cell communication clarifies how development and disease proceed, including cancer and inflammatory disorders, and supports the design of targeted therapies and tissue-engineering strategies.

Single-cell Microinjection for Cell Communication Analysis

We describe here how to perform a single-cell microinjection of Lucifer Yellow to visualize cellular communication via gap-junctions in living cells, and provide some useful tips. We expect that this paper will help everyone to evaluate the degree of cellular coupling due to functional gap junctions. Everything described here could be, in principle, adapted to other fluorescent dyes with molecular weight below 1,000 Daltons.

This video showcases a technique for establishing a co-culture of trigeminal neurons and primary dental pulp cells using a transwell filter. In this physically separated condition, mesenchymal cells obtained from dental pulp tissue secrete paracrine signaling molecules that traverse through the pores, promoting the differentiation and migration of neurons towards the mesenchymal cells establishing a co-culture system.

We adapted a permeable microporous membrane insert to mimic the tumor microenvironment (TME). The model consists of a mixed cell culture, allows simplified generation of highly enriched individual cell populations without using fluorescent tagging or cell sorting, and permits studying intercellular communication within the TME under normal or stress conditions.
