Epitope Retrieval

Epitope retrieval is a laboratory process that restores antibody-accessible sites, or epitopes, in biological specimens whose antigenic structures have been masked during fixation or sample preparation. In immunohistochemistry, heat-induced retrieval uses elevated temperature in a suitable buffer to disrupt formaldehyde-related cross-links, while enzyme-induced retrieval uses proteases to digest surrounding proteins and expose the target epitope. By improving antigen accessibility, the method strengthens antibody binding and signal detection in tissue sections and cell samples. Epitope retrieval is therefore essential for reliable localization and measurement of proteins in pathology, diagnostic research, developmental biology, and other microscopy-based studies.

SDS-PAGE/Immunoblot Detection of Aβ Multimers in Human Cortical Tissue Homogenates using Antigen-Epitope Retrieval

We describe a technique for the preparation of clarified human cortical homogenates, protein separation by SDS-PAGE, antigen retrieval and immunoblotting with an antibody to the Aβ peptide. Using this protocol, we consistently detect monomeric and multimeric Aβ in cortical tissue from humans with Alzheimer's pathology.

Peptide:MHC Tetramer-based Enrichment of Epitope-specific T cells

This protocol describes the use of peptide:MHC tetramers and magnetic microbeads to isolate low frequency populations of epitope-specific T cells and analyze them by flow cytometry. This method enables the direct study of endogenous T cell populations of interest from in vivo experimental systems.
