Live Cell Measurement

Live cell measurement is the quantitative analysis of living cells over time to assess their structure, behavior, and physiological state without stopping or destroying biological activity. It commonly uses time-lapse microscopy, fluorescent reporters, biosensors, or label-free imaging to track changes such as cell movement, division, morphology, intracellular signaling, and molecular dynamics under controlled conditions. In biology, these measurements reveal how cells respond to stimuli, communicate, migrate, differentiate, or undergo stress and death. By capturing dynamic processes rather than relying only on fixed end-point samples, live cell measurement supports studies of development, disease mechanisms, drug responses, and cell-based therapies.

2011

RNA polymerase II transcriptional kinetics are measured on specific genes in living cells. mRNAs transcribed from the gene of interest are fluorescently tagged and using Fluorescence Recovery After Photobleaching (FRAP) the in vivo kinetics of transcriptional elongation are obtained.

Several methods are available for measuring intracellular pH but very few of these allow simultaneous measurement of cytoplasmic and organellar pH. Here, we describe in detail a rapid and accurate methodology to simultaneously measure cytoplasmic and vesicular pH by ratiometric imaging of living cells.
